- _5 value "The GTP cyclohydrolase I (GTPCH) gene promoter consists of a Sp1/GC box, cAMP-response element (CRE), a CCAAT-box, and an E-box. Three distinct domains were detected in the promoter, Domain1 (+7 to -19, E-box), Domain2 (29-bp, -97 to -69, CAAT-box), Domain4 (-132 to -111, Sp1/Gc box). Promoter transcription was enhanced by 4-7 fold on incubation of PC12 cells with 8-bromo-cyclic AMP. Sp1/GC box sequence inhibited the transcriptional activity. The basal transcription was decreased by 66% on transfection of cells with the mutant CCAAT-box and CRE promoter. Both the CRE and CCAAT-box independently contribute to basal and cAMP-dependent activity. EMSA showed that nuclear proteins from PC12 cells bind the CCAAT-box as a complex containing NFY-B (Nuclear factor Y-B). The results thus demonstrate that NFYB binds to the CCAAT-box of the promoter and confers GTPCH transcriptional activation in response to cAMP." provenance.
