These animals had impaired tolerance to an i.p. injection of glucose (Fig. 2c), suggesting decreased insulin reserve, i.e., prediabetes. By 7 weeks of age, the blood glucose levels in Ncb5or -/- mice were 3-fold higher than those in +/+ mice during the fed state or after an overnight fast (Fig. 2a). The hyperglycemia in the Ncb5or -/- animals was associated with a marked reduction in plasma insulin. In the fed state, insulin levels of 7-week-old Ncb5or -/- animals were decreased to 25% of the levels in normal animals (Fig. 2b). Insulin was barely detectable in the plasma of fasting Ncb5or -/- animals (data not shown).
