- _5 value "Studies of the DBP gene proximal promoter have identified three functional hepatocyte nuclear factor 1 (HNF1)-binding sites within 2 kb of the transcriptional start site. The activities of the two more distal HNF1-binding sites were found to be dependent upon the relative levels of the two isoforms of this transcription factor, HNF1a and HNF1b. This was because HNF1b acted as a trans-dominant inhibitor of HNF1amediated enhancer activity14. The relative levels of these HNF1 isoforms in liver and kidney may contribute to the differences in the observed levels of DBP mRNA in these two tissues (Fig. 2). In contrast to their opposing effects on the DBP promoter, HNF1a and HNF1b both display only strong enhancing activities on the AFP and ALB promoters." provenance.
