Typically, an external signal or event eventually activates a MAP3K that then activates a MAP2K, which finally activates the MAPK (ERK, p38, JNK). In the p38MAPK branch, the directly relevant upstream kinases are MKK3 and MKK6, which appear to have redundant functions. Ask1, MEKK3 and Tak1 are amongst several possible MAP3K candidates for the p38MAPK branch, although these MAP3K often activate multiple MAPK pathways (reviewed in Kyriakis and Avruch, 2001).
